Analysis of Concentration and Purity of DNA Extraction on King Grass (Pennisetum purpuroides) and Elephant Grass (Pennisetum purpureum)

In line with the development of the livestock sector, the forage demand increases. Genetic improvements are needed through conventional techniques or through biotechnology which is currently developing at the molecular level. This study was conducted to determine the concentration and purity of DNA produced from king grass (Pennisetum purpuroides) and elephant grass (Pennisetum purpureum) using two DNA extraction buffers, namely CTAB and Kit DNAzol. This research used descriptive method. The observed variables were concentration and purity of DNA. The data were analyzed using descriptive statistical analysis. The results showed average concentration of king grass DNA using CTAB buffer and DNAzol buffer were 729.63 ng/ ?l and 142.72 ng/?l, respectively, and the average purity were 1.92 and 1.61. The average concentration of elephant grass DNA using CTAB buffer and DNAzol buffer were 44.8 ng/?l and 242.1625 ng/?l, respectively, and the purity were 3.05 and 1.8325. DNA extraction of king grass and elephant grass using the CTAB and DNAzol methods got varying results, the DNA extraction from king grass showed a better consentration and purity using CTAB buffer while elephant grass using DNAzol buffer.
